Copper tripeptide-1 and collagen research

GHK contains three amino-acid residues and binds copper(II). In a primary fibroblast-culture study, GHK-Cu increased collagen synthesis without increasing cell number. This distinction matters when interpreting a matrix-production endpoint: a change in collagen output is not necessarily a change in cell proliferation. Maquart and colleagues, FEBS Letters (1988).

In a separate rat wound-chamber model, investigators measured greater extracellular matrix accumulation and increased type I and type III collagen messenger RNA. These results identify experimental endpoints for matrix research; they do not establish a human outcome for this vial. Maquart and colleagues, Journal of Clinical Investigation (1993).

Extracellular matrix remodeling and turnover

Matrix remodeling includes both matrix production and breakdown. In cultured dermal fibroblasts, GHK-Cu increased matrix metalloproteinase-2 (MMP-2) expression and secretion, with effects on tissue inhibitors of metalloproteinases (TIMPs). Copper ions reproduced the MMP-2 effect in that study, while uncomplexed GHK did not. The result supports distinguishing the copper complex from GHK alone and measuring multiple matrix endpoints. Siméon and colleagues, Life Sciences (2000).
